Purificación parcial del compuesto proteico antimicrobiano por Paenibacillus Thiaminolyticus 165 en condiciones in vitro

Resumen
El objetivo de esta investigación fue caracterizar la bacteria Paenibacillus
thiaminolyticus 165, determinar su actividad antimicrobiana y purificar
parcialmente el compuesto antimicrobiano que produce. Se determinó la
actividad frente a bacterias gram positivas y gram negativas mediante la técnica
de difusión en agar. Posteriormente mediante cromatografía de intercambio
iónico se purificó el compuesto antimicrobiano. Los resultados indicaron que es
una bacteria Gram positiva con forma de bastón y generadora de esporas. Forma
colonias blancas, rugosas y con bordes rizoides en TSA. La bacteria presentó
crecimiento a pH neutro y a temperaturas de30o a 45o C. El péptido
antimicrobiano producido es activo frente a Salmonella spp., Escherichia coli,
Staphylococcus aureus, Listeria monocytogenes y Klebsiella spp. Las
condiciones óptimas para la mayor producción de péptidos antimicrobianos
tienen lugar en medio TSB (pH 7.1) a 30o C durante 27 horas. La técnica de
cromatografía de intercambio iónico utilizada no nos permitió una purificación del
compuesto antimicrobiano, debido probablemente a su naturaleza protéica o a
su conformación estructural.
This research was to characterize the bacterium Paenibacillus thiaminolyticus 165, determine its antimicrobial activity and partially purify the antimicrobial compound that produc. The activity against gram-positive and gram-negative bacteria was determined by the agar diffusion technique. Subsequently, by means of ion exchange chromatography, the antimicrobial compound was purified. Results indicated that is a Gram positive rod shaped bacterium and spore generator. It forms white, rough colonies with rhizoid borders on TSA. The bacteria presented growth at neutral pH and at a temperature of 45o C. The antimicrobial peptide produced is active against Salmonella spp., Escherichia coli, Staphylococcus aureus, Listeria monocytogenes and Klebsiella spp. The optimal conditions for the highest production of antimicrobial peptides take place in TSB medium (pH 7.1) at 30oC for 27 hours. Chromatography technique used did not allow a purification of the antimicrobial compound, probably due to its proteinic nature or its structural conformation.
