Excreción urinaria de Dipeptidil Peptidasa-4(DPP4) en un modelo experimental de Fibrosis

Resumen
[ES]El presente trabajo se centrará principalmente en el estudio de la relación entre la fibrosis renal, acumulación de colágeno en la matriz extracelular, y la actividad o cantidad de dipeptidil peptidasa-4 (DPP-4). Dicha enzima presenta actividad serina exopeptidasa que escinde los dipéptidos X-prolina a partir del extremo N-terminal de los polipéptidos. Debido a que el colágeno esta formado en su mayoría por residuos de glicina, prolina e hidroxiprolina, el depósito de colágeno en la matriz extracelular podría producir alteraciones en la expresión o en la actividad de esta enzima.
Para llevar a cabo el modelo de fibrosis renal, a un grupo de 10 ratas se le suministra durante 5 semanas una dosis de 2 mg/kg de cisplatino una vez/semana. El cisplatino es un nefrotóxico, ya que daña las células del epitelio tubular.
Una vez finalizado el experimento y analizado los resultados, obtenidos durante el desarrollo del mismo, se observó que la fibrosis renal modifica la excreción de la DPP-4 y por tanto podría ser útil como marcador de fibrosis renal, lo cual es de gran interés, tanto en el diagnóstico precoz como en el seguimiento de dicha patología.
[EN]This paper focuses primarily on the study of the relationship between renal fibrosis, collagen accumulation in the extracellular matrix, and the activity or amount of dipeptidyl peptidase-4 (DPP-4). This enzyme is an intrinsic membrane glycoprotein and has serine exopeptidase activity that rift the X-proline dipeptides from N-terminal end. Because of the collagen consists mainly of glycine residues, proline and hydroxyproline, collagen deposition in the extracellular matrix may produce alterations in the expression or in the enzyme activity. In order to carry out the renal fibrosis model, 10 rats were given 2 mg/kg of cisplatin for 5 weeks once a week. Cisplatin is a nephrotoxic, because it damages the cells of the tubular epithelium. Once the experiment was finished and the results have been analyzed, it shows that renal fibrosis effectively modifies the expression of the DPP-4, this could be useful like renal fibrosis marker, which is of great interest in early diagnosis and evaluation of such pathology.
