[ES]La enzima ARN polimerasa II de Saccharomyces cerevisiae (ARN pol II) se encuentra entre las polimerasas
eucarióticas mejor estudiadas. Esta enzima se compone de 12 subunidades que se ensamblan para formar un
complejo; 10 subunidades forman el core de la enzima, además del dímero disociable de las subunidades
rpb4 y Rpb7 (subcomplejo Rpb4/7). La transcripción por RNA pol II ocurre no sólo en genes codificantes de
proteínas anotados, sino en la mayoría del genoma y es fundamentalmente importante para la mayoría de
los procesos fisiológicos. La ARN pol II produce todos los ARNm y muchos ARN no codificantes. Existe un
equilibrio entre la transcripción y la degradación de los mRNAs, lo que implica la intervención de una
maquinaria compuesta por un enorme cantidad de proteínas. En el presente trabajo, estudiamos algunas
subunidades del ARN pol II y elementos proteicos de la maquinaria de degradación del ARNm. Esto se
realizó mediante técnicas de análisis de proteínas en dos cepas diferentes de la levadura S. cerevisiae; de tipo
silvestre y un mutante en el dominio conservado del pie de la ARN pol II denominado rpb1-84.
[EN]Saccharomyces cerevisiae RNA polymerase II (RNA pol II) enzyme is among the best studied of the eukaryotic RNA polymerases. RNA polymerase II is composed of 12 subunits; 10 subunit forms the Pol II core enzyme, in addition to a dissociable heterodimer of subunits Rpb4 and Rpb7 (Rpb4/7 subcomplex). The transcription by RNA pol II occurs not only at annotated protein-coding genes but mainly throughout the genome, and is fundamentally important for most physiological processes. RNA pol II produces all mRNAs and many noncoding RNAs. mRNAs balance between transcription and degradation, which imply the intervention of a machinery composed of a huge assembly of proteins. In the present work, we studied some subunits of RNA pol II, and proteins elements of the mRNA degradation machinery. This was done using protein analysis techniques in two different strains of the yeast S. cerevisiae; a wild type strain and a rpb1-84 mutant of the conserved domain of the foot of RNA pol II.
