Resumen
[ES] En estudios previos, se han aislado varias cepas de Lactobacillus pensotus a partir de la fermentación natural de la aceituna de mesa Aloreña. En este trabajo, hemos realizado ensayos para detectar la actividad antibacteriana de las diferentes cepas seleccionadas frente a Listeria sp. y Enterococcus faecalis S-47. Para ello, hemos ensayado dos técnicas diferentes utilizando como bacterias indicadoras diferentes cepas patógenas y vimos que casi todas las cepas producían sustancias antibacterianas en medio sólido pero no en medio líquido. Sin embargo, solo la cepa Lb. pentosus C-14 produjo sustancias antibacterianas en medio líquido, y esto determina que la mayoría de las bacteriocinas producidas por las cepas de Lb. pentosus ensayadas no se producen de forma constitutiva sino probablemente de forma inducida. La purificación de la bacteriocina producida por Lb. pentosus C-14 se llevó a cabo mediante cromatografía de intercambio catiónico ya que se trata de un péptido catiónico. El amplio espectro de acción de esta bacteriocina abarcó bacterias Gram-positivas y Gram-negativas, esto puede ser de gran interés ya que dicha cepa puede ser usada como cultivo iniciador y/o protector en varias fermentaciones o también como probiótico.
[EN] In previous studies, several strains of Lactobacillus pentosus have been isolated from the natural fermentation of Aloreña table olives. In this work, we performed tests to detect the antibacterial activity of the several selected strains against Listeria sp. and Enterococcus faecalis S-47. For this, we tested two different techniques using different pathogenic strains as indicator bacteria and we observed that almost all strains produced antibacterial substances in solid medium but not in broth. However, only Lb. pentosus C-14 strain produced antibacterial substances in broth, which suggest that most of bacteriocins produced by Lb. pentosus strains are not constitutively produced but probably under induction. Purification of bacteriocin produced by Lb. pentosus C-14 was carried out by cation exchange chromatography as it is a cationic peptide. The broad spectrum of action of this bacteriocin comprised Gram-positive and Gram-negative bacteria, this can be of great interest since this strain can be used as starter culture and/or protective culture in several fermentations or as a probiotic.
