Análisis de la interacción del lncRNA Gm35533 con la proteína Caveolina-3 y el microRNA-22 en el contexto cardiaco

[ES] Los RNAs no codificantes(ncRNAs) participan en la regulación de distintos procesos celulares. Estos
clasifican en RNAs no codificantes largos (lncRNAs >200nt) y RNAs pequeños (miRNAs ≈ 22nt). Los ncRNAs
interactúan con otros RNAs/proteínas inhibiendo su función uniéndose en la región 3’ UTR. En este estudio,
proponemos al lncRNA Gm35533 como regulador de la homeostasis en cardiomiocitos. Para conocer el
papel del lncRNA Gm35533, se estudia su interacción con el miR-22 y la Caveolina-3 en la pérdida de función
de este lncRNA, tratamiento con angiotensina II e isoproterenol en distintos tipos celulares. Nuestros resultados muestran que el lncRNA Gm35533 modula la expresión de la Caveolina-3 y miR-22, moléculas relacionadas con procesos de senescencia, migración y fibrosis celular. Concluimos que este lncRNA tiene un papel importante para mantener la homeostasis en cardiomiocitos, estableciendo una relación entre la Caveolina-3 y miR-22 mediante una regulación tejido específica.

These are classified into long non-coding RNAs (lncRNAs > 200 nt) and small RNAs (miRNA ≈ 22 nt). ncRNAs interact with other RNAs or proteins by inhibiting their function through recognition in the 3’UTR region. In this study, we propose lncRNA Gm35533 as a key regulator of homeostasis in cardiomyocytes. To understand the role of lncRNA Gm35533, we study the interaction between miR-22 and Caveolin-3 by loss-of-function assays, treatment with angiotensin II and isoproterenol in different cell types. Our results show that this lncRNA modulates the expression of Caveolin-3 and miR-22, molecules related to cardiomyocyte senescence, migration and cell fibrosis processes. We conclude that this lncRNA plays an important role in maintaning celular homeostasis in cardiomyocytes, establishing a relationship between Caveolin-3 and miR-22 through tissue-specific regulation.
