BIOMARCADORES COMUNES A LAS ENFERMEDADES GABAÉRGICAS Y SU APLICACIÓN A LA ENCEFALOPATÍA STXBP1

Abstract

[ES] El sistema GABAérgico presenta como función principal la inhibición sináptica. Dentro de este proceso, el gen STXBP1 se ve implicado en el acople de las vesículas sinápticas a la propia membrana sináptica, con el fin de liberar neurotransmisores inhibidores al medio sináptico, entre los que se incluye el GABA. Una de las alteraciones más graves en el desarrollo de la infancia viene determinada por mutaciones de novo en el gen STXBP1, lo que se denomina encefalopatía STXBP1. En este Trabajo de Fin de Grado se han obtenido las enfermedades que afectan al sistema GABAérgico, los genes comunes (biomarcadores) afectados en las mismas y los fármacos utilizados como tratamiento, para poder establecerlos como posibles terapias en la encefalopatía STXBP1, que ha sido posible gracias a la aplicación de la Biología de Sistemas.

[EN] The main function of the GABAergic system is the synaptic inhibition. STXBP1 gene regulates the coupling of the synaptic vesicles to the synaptic membrane, which results in the release of neurotransmitters to the synaptic cleft, including GABA. De novo mutations on the STXBP1 gene lead to the development of STXBP1 syndrome, which is characterised by a severe neurodevelopment alteration. In this study, the diseases that affect the GABAergic system have been identified, their common genes (biomarkers) that are affected among them and their target drugs, in order to introduce possible new therapies to deal with the STXBP1 encephalopathy under a Systems Biology approach.
