Resumen
[ES] Con este estudio se ha querido determinar el papel que desempeña el lncRNA Gm35533 durante el
desarrollo cardiaco embrionario. Pues a pesar de que se pensara en un principio, que los lncRNAs no tenían
ninguna función en especial, hay cada vez estudios más punteros que demuestran la gran importancia de
los ncRNAs. El estudio se ha realizado mediante el silenciamiento de dicho gen en un cultivo de células HL1,
seleccionadas debido a que son representativas del músculo cardiaco. El silenciamiento, se ha llevado a cabo
mediante una transfección en dichas células, usándose como molécula de interferencia un siRNA, ya que se
ha querido abordar inicialmente su estudio funcional en el citoplasma. Tras el procedimiento anterior, se ha
observado mediante inmunocitoquímica una disminución en MF-20, así como una bajada en los niveles de
ARNm de las miosinas mediante RT-qPCR, indicándonos una posible interacción del lncRNA Gm 35533 con
dichas proteínas.
[EN] The aim of this study was to determine the role of the lncRNA Gm35533 during embryonic heart development. Although it was initially thought that lncRNAs did not have any special function, there are more and more cutting-edge studies that demonstrate the great importance of ncRNAs. The study was carried out by silencing this gene in a culture of HL1 cells, selected because they are representative of cardiac muscle. Silencing was carried out by transfection in these cells, using an siRNA as an interference molecule, since the aim was to initially study its function in the cytoplasm. After the above procedure, a decrease in MF-20 was observed by immunocytochemistry, as well as a decrease in myosin mRNA levels by RT-qPCR, indicating a possible interaction of the lncRNA Gm 35533 with these proteins.
