Resumen
[ES] El cáncer es una de las enfermedades más letales en la actualidad, siendo la principal causa de muerte en los países desarrollados. En la actualidad existen numerosos tratamientos para combatir esta enfermedad, entre ellos el uso de fármacos, más conocido como quimioterapia. Uno de los principales objetivos en la terapia contra el cáncer es la inhibición de la enzima timidilato sintasa (TS) debido a la implicación de ésta en la síntesis del ADN de las células cancerosas. Se ha llevado a cabo una revisión bibliográfica de las características estructurales y funcionales de la enzima TS, así como de los inhibidores descritos para esta proteína folato dependiente. Esta revisión ha permitido realizar la proposición de diversas estructuras que puedan actuar como inhibidores de la enzima mediante su estudio a través de modelización molecular.
[EN] Cancer is one of the deadliest illness around the world, the main death cause in developed countries. Numerous types of treatment are currently available, including the use of drugs, better known as chemotherapy. One of the main targets in cancer therapy is the inhibition of the enzyme thymidylate synthase (TS) due to its involvement in DNA synthesis of cancerous cells. This work carries out a bibliographic review of the structural and functional characteristics of the TS enzyme as well as the inhibitors described for this folate-dependent protein. From the information retrieved, different structures that can act as enzyme inhibitors have been proposed and they have been studied by molecular modelling.
