Expresión, purificación, análisis funcional y mutación de c-Src

Abstract

[ES] Este trabajo estudia la proteína c-Src, un protooncogén clave en investigaciones sobre cáncer, biología estructural y fosforilación de quinasas. La sobreexpresión de c-Src, relacionada con la malignidad y resistencia en terapias oncológicas, se asocia con la agresividad tumoral y la supervivencia del paciente. En este estudio, se purificó, analizó funcionalmente y sometió a mutagénesis dirigida a c-Src, obteniendo un mutante para comparación funcional futura. Estos hallazgos podrían contribuir al desarrollo de inhibidores específicos para c-Src y sus variantes, con implicaciones en la innovación terapéutica.

[EN] This work explores the c-Src protein, a proto-oncogene integral to cancer research, structural biology, and kinase phosphorylation. Significant for its links to malignancy and resistance in cancer therapies, c- Src's overexpression correlates with tumor aggressiveness and patient survival. In this study, c-Src was successfully purified, analyzed functionally, and subjected to site-directed mutagenesis, resulting in a mutant for further functional comparison. These findings could facilitate the development of specific inhibitors targeting c-Src and its variants, with implications in therapeutic innovation.
