Resumen
[ES] La neuroglobina (Ngb) es una proteína de 17KDa perteneciente a la familia de las globinas. Se encuentra en todos los vertebrados y se expresa en diversos tejidos, pero muestra una ubicación preferencial en el sistema nervioso. Su función específica se desconoce, sin embargo, se ha demostrado que la Ngb es una molécula endógena con acción neuroprotectora, de ahí el interés en su empleo para el tratamiento patologías como la isquemia. Con este fin, se ha llevado a cabo la purificación de la Ngb de rata, utilizando un sistema de expresión de proteínas de fusión con el empleo del plásmido pET15b y posterior trasformación en la cepa de E.coli BL21(DE3). Esto ha permitido la expresión de la proteína con una cola de histidina, facilitando su unión a una resina de afinidad por unión a metal. El seguimiento de este protocolo de purificación ha permitido la obtención de Ngb con alto grado de pureza.
[EN] Neuroglobin (Ngb) is a 17KDa protein belonging to the globin family. lt is found in ali vertebrates and is expressed in various tissues, but it shows a preferential localization in the nervous system. lts specific function is unknown, however, accumulating evidence hace clearly demonstrated that Ngb is an endogenous neuroprotective molecule, hence the interest in its use for the treatment of pathologies such as ischemia. To this end, the purification of rat Ngb was carried out, using a fusion protein expression system with the use of the plasmid pET15b and subsequent transformation in E.co/i strain BL21 (DE3). This has allowed the expression of the protein with a histidine tag, favoring its binding to an affinity resin by metal binding. The monitoring of this purification protocol has allowed to obtain the Ngb with a high degree of purity.
