Cultivo in vitro de Gimnospermas

Abstract

[ES] El presente trabajo tiene como objetivo determinar la influencia del medio de cultivo, condiciones de iluminación y vernalización sobre el cultivo in vitro de cinco coníferas y, concretamente, sobre la germinación, inducción de callo y embriogénesis somática. El mejor explante para la inducción de callo fue el cotiledón y que las condiciones idóneas de su cultivo fueron el uso del medio MS en condiciones de iluminación. En la etapa de proliferación, el cotiledón dio lugar al mayor crecimiento de las masas embrionarias, siendo la luz y el medio LP las mejores condiciones de cultivo. En el caso de la germinación y la medida de los parámetros de crecimiento, ni la vernalización ni el tipo de medio de cultivo influyeron sobre ambos, solo en el caso de Platycladus orientalis, en el cual la vernalización favoreció la germinación y el medio MS incrementó la longitud total y de la raíz.[EN] The present work aim to determine the influence of the type of culture medium, lighting conditions and vernalization on the in vitro culture of five conifers and, specifically, on germination, callus induction and somatic embryogenesis. It was determined that the best explant for callus induction was cotyledon and the ideal conditions for its cultivation were the use of MS medium under lighting conditions. In proliferation stage, cotyledon increased growth of embryonic masses, being light and LP medium the best culture conditions. In the case of germination and measurement of growth parameters, neither vernalization nor the type of culture medium influenced both, only in the case of Platycladus orientalis, in which vernalization benefited germination and MS medium increased the total and root length of the plants.
