[ES]La transcripción es un proceso regido por la actividad de las RNA polimerasas. En concreto, la RNA pol II se encarga de transcribir los mRNAs. Esta enzima se compone de 12 subunidades que se ensamblan para formar un complejo. Dos de esas subunidades se disocian en forma de heterodímero en determinados momentos de la transcripción, lo cual parece tener una función importante en la vida de los mRNAs. El objetivo de este trabajo ha sido estudiar diferentes subunidades de la RNA pol II, de forma que se analice el ensamblaje de la enzima, así como a las proteínas que intervienen regulando los niveles de mRNA (transcripción y degradación del mRNA), en cepas mutantes de la transcripción. La metodología empleada en el seguimiento de los objetivos ha consistido en una novedosa técnica de purificación de cromatina, seguida de análisis de proteína por western-blot. Además, a partir de los western-blot se han hecho cuantificaciones por densitometría en las diferentes situaciones experimentales. Los resultados obtenidos revelan que la asociación de Rpb4 a la cromatina es dependiente de RNA en una cepa silvestre. Además de Rpb4, que interviene en el denominado Cross-talk del mRNA, otras dos proteínas implicadas en dicho proceso, Not4 y Xrn 1, parecen estar asociadas a la cromatina de forma dependiente de RNA. Dichas proteínas también se ven afectadas en los análisis en una cepa rpb44, por lo que es importante analizar en conjunto estas proteínas, ya que parece que tienen mucha importancia en la regulación de la estabilidad y la vida media de los mRNAs.
[EN]Transcription is a process governed by the activity of RNA polymerases. Specifically, RNA pol II is responsible for transcribing the mRNAs. This enzyme is composed of 12 subunits that are assembled to form a complex. Two of these subunits dissociate in the form of heterodimer at certain moments of transcription, which seems to have an important function in the life of the mRNAs. The objective of this work is to study different subunits of RNA pol II, in order to analyze the assembly of the enzyme, as well as the proteins that regulate mRNA levels (mRNA transcription and degradation), in mutant strains of the transcription. The methodology used in this work consisted of a novel technique of chromatin purification, followed by protein analysis by western-blot. In addition, quantification by densitometry in the different experimental situations has been made from the western blot. The results obtained reveal that the association of Rpb4 to chromatin is dependent on RNA in a wild strain. In addition to Rpb4, which is involved in the so called mRNA Cross-talk, two other proteins involved in this process, Not4 and Xrn1, appear to be associated with chromatin in an RNA-dependent manner. These proteins are also affected in analyzes in a rpb4A strain, so it is important to analyze these proteins together, since they seem to be very important in regulating the stability and half-life of mRNAs.
