EFECTO DEL TIROSOL EN LA RESPUESTA DE LOS FACTORES DE TRANSCRIPCIÓN INDUCIBLES POR HIPOXIA

Resumen
[ES]En los tumores sólidos se generan ambientes hipóxicos en los que se expresan factores de
transcripción como HIF-1 y HIF-2, asociados con un mal pronóstico. El consumo de aceite de
oliva se ha vinculado con una reducción del riesgo a padecer enfermedades crónicas y
determinados tipos de cáncer, como el cáncer de mama. Dado que el tirosol (Tir) es uno de sus
principales compuestos fenólicos, en este TFM se evaluó su efecto sobre la expresión y actividad
tanto de HIF-1 y HIF-2 en una situación de hipoxia en la línea celular de cáncer de mama MCF-
7. Los resultados muestran que el tratamiento con Tir a concentraciones de 5, 10 y 200 μM
disminuye el nivel de proteína HIF-1a, y a una concentración de 75 y 200 μM el de HIF-2a, sin
afectar en ningún caso la expresión de su ARNm. También se observó que el Tir no modifica la
expresión de los genes diana de HIF-1 y HIF-2 analizados. Sin embargo, sí ejerce un efecto sobre
el ciclo celular que podría estar relacionado con una mayor actividad de HIF-2. Concretamente
favorece la proliferación celular al ocasionar una disminución de células en fase G0/G1 y un
aumento en la fase S a concentraciones de 75, 100 y 200 μM. En definitiva, nuestros resultados,
aunque preliminares, no permiten afirmar que el Tir ejerza un efecto en la respuesta de HIF-1 y
HIF-2 en células MCF-7 sometidas a hipoxia.

[EN]Hypoxic regions are inherent to solid tumors. In those regions there is an induction of HIF-1 and HIF-2, two transcription factors associated with poor prognosis. The intake of olive oil has been linked to a lower risk of chronic diseases and of certain types of cancer, such as breast cancer. Given that tyrosol (Tyr) is one of the main phenolic compounds in olive oil, we have evaluated its effect on the expression and activity of HIF-1 and HIF-2 in hypoxic MCF-7 breast cancer cells. Our results show that treatment with Tir at 5, 10 and 200 μM decreases the protein level of HIF-1α and at 75 and 200 μM downregulates HIF-2α without affecting, in any case, their mRNA expression. Moreover, Tyr doesn´t influence the expression of HIF-1 or HIF-2 target analyzed. However, it does exert an effect on the cell cycle that could be related to a higher activity of HIF-2. Particularly, at concentrations of 75, 100 and 200 μM, it seems to favor cellular proliferation by decreasing the percentage of in cells in G0/G1 phase and increasing the percentage of cells in S phase. In conclusion, our results, although preliminary, do not allow us to affirm that Tyr can modulate the response of HIF-1 and HIF-2 in MCF-7 hypoxic cells.
